I am Using P-gal4 mediated enhncer trap system to Identify genes exprssed in drosophila devlopment. I am using the self ligated Plasmids with P-Gal4 along with Few genomic sequences.

  1. I hope I am understanding this question correctly, but if you are truly performing inverse PCR with self-ligated plasmids, wouldn’t you need the reverse compliments of those primers? Also, if I’m not mistaken, in inverse PCR since you are randomly cutting the vectors isnt’ there also a chance you won’t have those sequences in the final ligated product?
    Whenever I did inverse PCR I used a reverse compliment of an interior sequenc of interest and a reverse compliment of a vector sequence.
